Method Autotrophic Growth Conditions. A 1-L culture of Synechocystis was cultivated autotrophically in a 1.25-L bioreactor (Bioflo 3000, New Brunswick Scientific, NJ) under a surface light flux of 150 µE/m^2/s in BG-11 medium (American Type Culture Collection medium 616) buffered with 10 mM HEPES under atmospheric levels of carbon dioxide at 30°C. The pH was controlled at 7.5±0.1. Growth was monitored spectrophotometrically by measuring the optical absorbance of cultures at 600 and 730 nm.
Comments The nucleic acids were divided into 17% RNA and 3% DNA constituents (Neidhardt, F. C. Ingraham, J. L. Schaechter, M. Physiology of the Bacterial Cell. A Molecular Approach Sinauer Associates, Inc.: Sunderland, MA, 1990.) The final carbon and nitrogen balances were within 1.8% and 6.5% of the elemental analysis (see BNID 105530).
