Reference Bult et al., Complete genome sequence of the methanogenic archaeon, Methanococcus jannaschii. Science. 1996 Aug 23 273(5278):1058-73. p.1058 right-most column bottom paragraph.PubMed ID8688087
Method p.1058 right column 2nd paragraph:"A whole-genome random sequencing method (Fleischmann et al., 1995 PMID 7542800 and Fraser et al., 1995 PMID 7569993) was used to obtain the complete genome sequence for M. jannaschii A small insert plasmid library [average insert size, 2.5 kilobasepairs(kbp)] and a large-insert ? library (average insert size, 16 kbp) were used as substrates for sequencing. The ? library was used to form a genome scaffold and to verify the orientation and integrity of the contigs formed from the assembly of sequences from the plasmid library."
Comments "The M. jannaschii genome consists of three physically distinct elements: (i) a large circular chromosome of 1,664,976 base pairs (bp) (Fig. 1), which contains 1682 predicted protein-coding regions and has a G+C content of 31.4% (ii) a large circular extrachromosomal element (ECE) (H. Zhao et al., Arch. Microbiol. 150,178 (1988)) of 58,407 bp, which contains 44 predicted protein-coding regions and has a G+C content of 28.2% (Fig. 2) and (iii) a small circular ECE (H. Zhao et al., Arch. Microbiol. 150,178 (1988)) of 16,550 bp, which contains 12 predicted protein-coding regions and has a G+C content of 28.8% (Fig.2)" Total genome size=1,739,933bp. Total gene number=1,682+44+12=1,738genes
