Method A streptomycin-resistant mutant of H. influenzae type b strain Rd/b+, a laboratory transformant, was used because it is highly virulent in rats. Bacteria were grown to mid-logarithmic phase in supplemented brain heart infusion broth and inoculated into rats that had undergone splenectomy or a sham operation.
