Method As a first step to examining polymerase fidelity, the rates of incorporation of correct nucleotides were examined for all four dNTPs, utilizing a template containing the appropriately matched base. Pre-equilibrated polymerase-DNA complex was mixed with various concentrations of dNTP to examine formation of correct base pairs. The rates of the pre-steady state bursts were plotted as a function of dNTP concentration and fit to a hyperbola (burst rate =k pol[dNTP]/(Ka + [dNTP])) to yield the Ka and maximum polymerization rate, k pol, for each nucleotide incorporation.
