Method The yeast is first cultivated for 4 h in a complete Rider medium containing 9 mM Pi and 111 mM glucose as a carbon source. Then the cells are placed on the same medium except depleted of Pi under sterile conditions and the culture is grown during 7 h. Further, the cells are again placed on fresh complete medium, where the culture is growing for 2 and 4 h
Comments The polyP3 fraction is involved in the maintenance of the fine structural organization of the yeast cell wall. This suggests that the cells primarily restore normal function of the cell surface disrupted during the phosphate starvation.
