Method Chromosomal localization of guinea pig kininogen gene observed by fluorescence in situ hybridization (FISH) analysis. Guinea pig cardiac blood lymphocytes were cultured in the presence of phytohemagglutinin to be introduced into the cell proliferation cycle. The prometaphase chromosome spreads of the proliferating lymphocytes were hybridized with a fluorescent-labeled DNA probe that was a long polymerase chain reaction product with a 5-kbp length of the guinea pig kininogen gene. The guinea pig chromosome number counted was 64 (2n), which is consistent with the previous reports [refs 18, 19 in reference].
