In arabinose system in e. coli. Translation was blocked by the addition of 200 µg/ml chloramphenicol, 30 min after the induction of gfp-expression with 0.2% arabinose. Fluorescence images were acquired every 3–5 min before and after inhibition.The statistics was obtained from 77 cells. |

All FPs, including derivatives of Aequorea victoria GFP, require a posttranslational reaction to form the fluorophore. For GFP derivatives, fluorophore maturation proceeds through two sequential steps: a relatively fast cyclization of the three amino acids at residues 65-67 followed by a slow oxidation step. This slow step dominates the maturation kinetics, and chromophore maturation effectively follows first order kinetics. |
