Measurement MethodPlant growth. Arabidopsis thaliana var Col-0 was grown in soil [GS 90 soil mixed with vermiculite in a ratio 2:1 (v/v)]. After germination the seedlings were grown for 1 week in a 16/8 h light (250 µE m−2 s−1, 20 °C)/dark (6 °C) regime, for 1 week in an 8 h light (160 µE m−2 s−1, 20 °C)/16 h dark (16 °C) regime, and then replanted with one seedling per pot and transferred for 1 week to growth cabinets with an 8/16 h light–dark cycle (160 µE m−2 s−1, 20 °C throughout the day/night cycle). The plants were then distributed into small growth cabinets with a 12, 8, 4, 3 or 2 h photoperiod (all with a light intensity of 160 µE m−2 s−1 and 20 °C throughout the day/night cycle). The plants were harvested 17 d later. Typically, at least five separate samples, each containing three rosettes, were harvested. Extraction and assay of enzymes.
