Method Pre-cultures were grown overnight in 1,000 ml shake-flasks filled with 100 ml medium (37°C, 250 rpm).
Comments As the adenylates energy charge is computed as: EC=([ATP]+[ADP]/2)/([ATP]+[ADP]+[AMP]) there would be no influence on the EC in case of unspecific leakage (e.g., 70% loss of ATP, ADP and AMP). Using the corrected values of adenylates found in this study the EC is between 0.72 (quenching fluid H) and 0.59 (quenching fluid G). Quenching fluid G=60% (v/v) methanol/H2O with 30 mM TEA.
