Method Escherichia coli was exposed to partial pyrimidine starvation by feeding a pyrBI strain orotate as the only pyrimidine source. Ribosome synthesis rate was determined according to prokaryotic elongation factors (EF-Tu (elongation factor thermo-unstable)) and ribosomal protein S1): Alpha was determined by growing the cells with [3H]lysine and pulse-labeling for 2 min with [35S]methionine.
Comments Table gives rRNA synthesis rates (Alpha (EF-Tu or S1) means the differential rate of synthesis of that particular protein relative to the rate of total protein synthesis in arbitrary units for each protein), DNA, RNA, protein, ppGpp (Guanosine tetraphosphate), and nucleoside triphosphate levels for E. coli grown on different levels of pyrimidine supply with differing Doubling time. The nucleotide pools are expressed as picomoles per OD436 unit.
