Comments Abstract: "In contrast, no changes were observed in phospholipid amount, in phospholipid composition, and in phosphatidylethanolamine plasmalogen content during the entire disease process. The fatty acid and aldehyde composition of individual phospholipids remained unaltered as well. No modifications could be detected in cholesterol content." P.282 right column 3rd paragraph: "The phospholipid pattern, including the alkenylacyl-PE/diacyl-PE ratio, was also unaffected by the disease. The fatty acid and aldehyde composition of the individual phospholipid classes remained similarly unmodified. Furthermore, cholesterol, which constitutes one-third of total brain lipid amount, exhibited no changes during the whole disease process." P.283 right column bottom paragraph: "The modifications in the levels of mevalonate pathway lipids (BNID 103293, 103294, 103295, 103296) in scrapie-infected mouse brain point to several possible key events in the disease process. These include modified protein glycosylation, changes in the synthesis of GPI anchors, and increased production of free radicals. This is in great contrast to the stability of the phospholipids and cholesterol. Thus, scrapie infection affects specific parts of lipid metabometabolism and does not induce general membrane destruction in brain through lysosomal breakage."
