Method P.2105 left column 3rd paragraph: "Translation was blocked by the addition of 200 µg/ml chloramphenicol, 30 min after the induction of gfp-expression with 0.2% arabinose. Fluorescence images were acquired every 3–5 min before and after inhibition. As this measurement was more sensitive, the illumination was reduced and the EM gain of the camera was used. Photobleaching could thus again be neglected. Cellular fluorescence was determined by summing all pixel values above the background level for each bacterium. This method is qualitatively equal to the use of cell outlines as described above, but can only be applied if the range of fluorescence values is limited and bacteria do not grow strongly. The resulting maturation time courses were fitted by an exponential function."
Comments P.2106 right column bottom of paragraph: "The distribution of time-constants τm of GFP maturation shown in Fig. 4 b was obtained from exponential fits to 77 single-cell time series (solid lines in Fig. 4 a). [Investigators] find an average maturation time of τm = 6.5 min and a standard deviation of 0.6 min, i.e., a cell-to-cell variation of only ~10%."
