Each MA line was grown on YPD solid medium (1% yeast extract, 2% peptone, 2% dextrose, and 2% agar) at 30° and passaged by single-cell transfer." P.1818 right column 2nd paragraph: "[Researchers] estimated the average number of generations assuming exponential growth from counts of the number of cells per colony. The number of cells per colony was estimated approximately every seven transfers by choosing a single colony from each of 10 petri dishes. [They] then suspended the colony in 1 ml of water and estimated cell density using a hemacytometer. (Reichert Bright Line, 0.1 mm depth)." |

YPD Medium is a blend of peptone, yeast extract, and dextrose in optimal proportions for growing most Saccharomyces cerevisiae strains. |
