Method “To determine how Ca2?+/calcineurin-dependent modulation of phosphorylation at these sites [seven phosphorylation sites in Kv2.1] regulates voltage-dependent gating of Kv2.1, [researchers] expressed isoforms carrying mutated phosphorylation sites and wild-type Kv2.1 in HEK293 cells [Human Embryonic Kidney 293]. Whole-cell patch-clamp was used to analyze Kv2.1 in untreated cells, and cells treated with ionomycin, or with AP dialyzed through the patch pipette.”
